Native GHRH arrives at the pituitary, activates the receptor, and is destroyed almost immediately. DPP-IV cleaves it within minutes. The signal is real but too short to produce meaningful GH output. CJC-1295 without DAC (Modified GRF 1-29) is a synthetic 29-amino acid analog of GHRH developed through systematic structural modification of Sermorelin. The core insight: Sermorelin's limitation was never potency. It was survival. Native GHRH is destroyed within minutes by serum proteases. Four targeted amino acid substitutions protect the peptide from enzymatic cleavage without altering receptor interaction. Position 2: D-alanine blocks DPP-IV cleavage. Position 8: glutamine prevents deamidation. Position 15: alanine stabilizes a secondary cleavage site. Position 27: leucine eliminates oxidation-vulnerable methionine. The result is an effective half-life of approximately 30 minutes versus 5-10 minutes for native GHRH. Same receptor. Same pituitary pathway. Same pulsatile release. Three to six times the duration.
GH secretion increases through both immediate granule release and enhanced GH gene transcription, building pituitary reserve over time. IGF-1 elevation follows proportionally, supporting protein synthesis, fat metabolism, and tissue repair. Combined with Ipamorelin, CJC-1295 forms the most prescribed peptide pairing in GH optimization: dual-receptor activation (GHRH + ghrelin pathways) converging on the same somatotroph cell for synergistic output.
Administer via subcutaneous injection using a 29-31 gauge insulin syringe. Inject before bed. Abdomen or upper thigh preferred.
