- Semax is a synthetic heptapeptide derived from the 4-10 fragment of adrenocorticotropic hormone (ACTH). Developed in the 1980s at the Institute of Molecular Genetics of the Russian Academy of Sciences, it has been approved as a pharmaceutical in Russia since 1996 for stroke, cognitive disorders, and optic nerve disease.
- Native ACTH(4-10) degrades within 30-60 minutes. The addition of a C-terminal Pro-Gly-Pro tripeptide extends biological duration to 20-24 hours while keeping the molecule at 813.92 Daltons, small enough to cross the blood-brain barrier efficiently after subcutaneous injection.
- Semax operates through four parallel pathways: BDNF/TrkB upregulation in the hippocampus, serotonergic activation, dopaminergic modulation for executive function, and enkephalin preservation for stress resilience. Despite ACTH derivation, it has zero hormonal activity. No cortisol stimulation. No adrenal axis disruption.
- BDNF is the master regulator of neuroplasticity, governing neuronal survival and the long-term potentiation underlying learning and memory. A single dose of Semax produced a 3-fold increase in BDNF gene transcription, 1.4x increase in BDNF protein, and 2x increase in TrkB receptor mRNA. The brain both produces more BDNF and becomes more responsive to it.
- Beyond BDNF, Semax potentiates dopaminergic signaling in prefrontal circuits without forcing dopamine release. Genome-wide analysis shows it modulates over 1,500 genes, selectively activating neurotrophin transcription in stressed neural tissue.
Administer via subcutaneous injection into the abdomen and rotate injection sites with each dose to help minimize skin irritation.
